Abstract Background Our previous studies reveal that impaired draining function of the synovial lymphatic vessel (LV) contributes to pathogenesis inflammatory arthritis, but cellular and molecular mechanisms involved are not fully understood. Objective To investigate involvement muscle cells (LMCs) in mediating LV arthritis. Methods TNF transgenic (TNF-Tg) arthritic mice were used. The structure LVs drained hind limbs examined by whole-mount immunofluorescence staining, electron microscopy, near-infrared imaging. Primary LMCs treated with TNF, changes proliferation, apoptosis, functional gene expression assessed. roles herbal drug, Panax notoginseng saponins (PNS), arthritis studied. Results TNF-Tg developed ankle age, which was associated abnormalities LVs: (1) dilated capillary few branch points, (2) mature reduced LMC coverage function, (3) degenerative apoptotic appearance LMCs. caused genes, promoted production nitric oxide (NO) endothelial (LECs). PNS attenuated restored LVs, inhibited TNF-mediated NO expression, apoptosis.
